Determinación sexual de aves mediante métodos moleculares

Abstract

[Resumen]: En el presente TFG, los primers P2/P8 propuestos por Griffiths et al. (1998) se han utilizado para tratar de sexar muestras de mosquitero ibérico (Phylloscopus ibericus). Se han encontrado una serie de dificultades motivadas por la antigüedad de algunas de las muestras. Una vez superadas, aparecen nuevos problemas derivados del uso de los primers anteriormente mencionados combinados con el uso de gel de agarosa. Para tratar de aportar luz se toman una serie de muestras de aves domésticas, entre ellas de gallinas de la variedad Brahma (Gallus gallus domesticus) y faisán común (Phasianus colchicus), y se tratan de sexar, obteniendo los mismos resultados. Finalmente, tras una búsqueda bibliográfica se llega a la causa, que no es otra que el uso de primers que aportan muy poca diferencia entre las secuencias amplificadas.[Abstract]: In the present TFG, the P2/P8 primers proposed by Griffiths et al. (1998) have been used to treat to determine the sex of iberian chiffchaff samples (Phylloscopus ibericus). A series of difficulties have been found due to the age of some of the samples. Once overcome, new problems arise from the use of the previously mentioned primers combined with the use of the agarose gel. In order to shed some light on, a series of samples of domestic birds, hens of the Brahma variety (Gallus gallus domesticus) and common pheasant (Phasianus colchicus) are taken and tried to determine sex, obtaining the same results. Finally, after a bibliographic search, the cause is reached, which is nothing else than the use of primers that provide very little difference between the amplified sequences.
